چکیده
Aim of this work is to present the phenotype and biochemical characteristics primary freshly isolated murine hepatocytes exposed oleic palmitic acids, most abundant fatty acids (FAs) in human diet serum. Excess FAs one factors that contributes non-alcoholic liver disease (NAFLD). The hallmark NAFLD overaccumulation lipids hepatic tissue formation lipid droplets (LDs). With time can progress into more advanced stages leading injury. Hepatocytes as cells are strongly involved process LDs formation, therefore widely used research on molecular mechanism development progression. Primary rarely investigations, due multistep isolation loss functionality under culture conditions. However, they offer substantial advantages, including preserved functions. In order investigate their content, Raman FTIR imaging were employed, provide spatial vibrational information studied label-free manner.

Pretranslational regulation of cytochrome P4504A1 by free fatty acids in primary cultures of rat hepatocytes.
The effect of different fatty acids on cytochrome P4504A1 mRNA levels was studied in primary cultures of rat hepatocytes, using a solution hybridization assay. All fatty acids tested induced P4504A1 mRNA levels in a dose- and time-dependent manner. Most potent were docosahexaenoic acid (22:6) and arachidonic acid (20:4), both of which gave a 6-fold increase in mRNA levels at 300 microM, followe...
متن کاملPAT protein mRNA expression in primary rat hepatocytes: Effects of exposure to fatty acids.
Excess energy is stored as neutral lipids in lipid droplets (LDs) whose surface is coated by PAT proteins, each playing a distinct cellular function. The adipocyte differentiation-related protein (ADRP) and tail-interacting protein (TIP47) are expressed almost ubiquitously, whereas the oxidative tissue-enriched PAT protein (OXPAT) is expressed in specific tissues, such as the liver. In rat live...
متن کاملBiodegradable Starch - Based Films Containing Saturated Fatty Acids : Thermal , Infrared And Raman Spectroscopic Characterization
Biodegradable films of thermoplastic starch and poly (butylene adipate co-terephthalate) (PBAT) containing fatty acids were characterized thermally and with infrared and Raman spectroscopies. The symmetrical character of the benzene ring in PBAT provided a means to illustrate the difference between these spectroscopic techniques, because a band appeared in the Raman spectrum but not in the infr...
متن کاملMonitoring cellular responses upon fatty acid exposure by Fourier transform infrared spectroscopy and Raman spectroscopy.
We investigated the applicability of FTIR-spectroscopy as a high throughput screening method for detection of biochemical changes in intact liver cells in bulk upon fatty acid exposure. HepG2 cells adapted to serum free (HepG2-SF) growth were exposed to four different fatty acids, three octadecenoic acids, differing in cis/trans-configuration or double bond position (oleic acid, elaidic acid an...
متن کاملUrsodeoxycholyl lysophosphatidylethanolamide inhibits lipoapoptosis by shifting fatty acid pools toward monosaturated and polyunsaturated fatty acids in mouse hepatocytes.
Ursodeoxycholyl lysophosphatidylethanolamide (UDCA-LPE) is a hepatoprotectant in inhibiting apoptosis, inflammation, and hyperlipidemia in mouse models of nonalcoholic steatohepatitis (NASH). We studied the ability of UDCA-LPE to inhibit palmitate (Pal)-induced apoptosis in primary hepatocytes and delineate cytoprotective mechanisms. We showed that lipoprotection by UDCA-LPE was mediated by cAM...
